ヒト気道アレルギー性炎症モデルによる炎症細胞ホーミング機構の解析:SCIDマウスを用いた喘息モデル作成と治療の研究

研究概要	肺癌による手術にて得られた肉眼的に正常な気管支組織をSCIDマウスの皮下に移植すると、SCIDマウスと移植ヒト気管支の血管が交通して血流が得られることが確認された。そこで、アトピー型喘息患者の末梢血単核球をSCIDマウスの腹腔内に移注すると移植ヒト気管支組織に喘息患者の末梢血単核球が浸潤することが確認された。移植ヒト気管支組織に浸潤したリンパ球を分析すると、喘息患者の末梢血単核球を移駐した群では、健常者の末梢血単核球を移駐した群より有意にCD3陽性細胞が増加しており、その中でCD4陽性細胞数が有意に多かった。さらに、気道に遊走したリンパ球の中では接着分子であるCD103(αeβ7 integrin)陽性細胞が有意に多かった。これらの結果から、喘息の発症機構の一つに喘息患者のリンパ球が気道組織へ移行しやすい特性(ホーミング機能)を持ち、気道局所で抗原提示細胞とリンパ球が炎症の中心的な機能している可能性が示唆される。今後、アトピー型喘息患者の末梢血単核球を移注したSCIDマウスの移植気道を利用して、受動免疫を介するアレルギー反応を誘発して、炎症細胞を遊走させ、そこで慢性のアレルギー性炎症を誘導する計画である。これらの研究に加えて、私達のグループが初めて確認したヒト気管支上皮内リンパ球の特性を検討した結果、気管支上皮内リンパ球は、上皮下リンパ球と比較して、活性化リンパ球は少ないが、CD103(αeβ7 integrin)が強く発現する事を確認した。また、喘息患者の気管支上皮内リンパ球は、非喘息患者のものと比較してCD4が少なく、CD8が多いことも判明した。しかしながら、まだ、これらの機能的意義については説明出来ないので今後検討を続けていく予定である。
