アレルギーにおけるT細胞受容体と抗原提示分子の構造と結合に関する研究

研究概要	(1)抗原の特性 β-lactoglobulin(BLG)のoverlapping peptidesを合成して抗原として用い、複数の抗原特異的T細胞を樹立した。BLGには抗原特異的T細胞によって認識されやすい部位があり、最も認識されやすい抗原決定基はBLGp101-111(KYLLFCMENSA)であった。 (2)抗原提示分子の特性牛乳アレルギー群とコントロール群でのHLADRB10405の保有率の間に有意差は認められなかったが、BLGの抗原提示分子としてDRB10405の有意な関与が示された。 (3)T細胞受容体(TCR)の特性 BLG抗原特異的T細胞のTCR usageに関しては一定の傾向が認められなかったが、CDR3領域には類似の性質を持つアミノ酸配列が認められた。 (4)サイトカイン産生 BLG抗原特異的T細胞株では大量のIL-5産生がすべての細胞株で認められ、IL-4産生量は個々の細胞株で同等であったがIFN-γの産生量に差が認められ、IL-4/IFN-γ比は即時型で増加し非即時型では減少したことから、Th1・Th2バランスの傾きによって異なる病態が形成されると考えられた。 (5)アナログペプチドに対するT細胞応答 BLGp101-111のアミノ酸を1つづつ別のアミノ酸に置換して合成したアナログペプチドに対するT細胞クローンの反応性の検討では、類似の性質を有するアミノ酸に置換した場合にのみT細胞応答が認められたことから、抗原のもつ生化学的特性がアレルギー病態におけるT細胞応答を規定すると考えられた。Th2型からTh1型への異なるT細胞応答の誘導によるアナログペプチド療法の開発を目指している。
