細胞接着ならびに細胞増殖に関わるシグナル伝達への放射線照射の影響に関する研究

研究概要

p53がん抑制遺伝子(p53)に代表されるDNA損傷を起点とする細胞死のシグナルだけでなく、細胞膜上の受容体を起点とする生存シグナル伝達の活性化が抗癌剤や放射線に対する感受性の決定要因として重要であると考えられることから,野性型p53ならびに変異型p53を有する2種類のヒト食道扁平上皮癌細胞株を用いてTyrosine kinase InhibitorであるGenisteinの放射線感受性の修飾効果を生存シグナルに焦点を充てて検索した.照射とGenisteinを併用すると生存シグナルであるp42/p44 extracellular signal-regulated kinase(ERK)とAKT/PKBの照射による活性化は抑制され,放射線感受性が両細胞ともに増強した.また,野性型p53を有する腫瘍細胞株ではBaxの上昇,Bcl-2の減少ならびにPARPの分解が認められ,アポトーシスが誘導されたが,変異型p53を有する細胞株ではp53関連蛋白の変動ならびにアポトーシスの誘導は認められなかった.したがって,照射とGenisteinの併用によって誘導される細胞死の機序は野性型p53と変異型p53を有する細胞では異なることが明らかとなった.また,ERKやAKT/PKBを含む生存シグナルは放射線感受性の決定に関与しており,生存シグナルを阻害するGenisteinは放射線感受性を高める可能性をもった薬剤となりうることが示唆された.
