研究概要	「非アルツハイマー型変性痴呆の病態機序に関する研究-細胞骨格蛋白異常の観点から」のうち、最終年度は主としてレビー小体型痴呆(DLB)の細胞病理学的研究を行った。まず、レビー小体の神経細胞における形成過程を明らかにするため、α-シヌクレイン免疫組織化学を用いて、レビー小体の形成を6段階に分類してその超微形態の特徴を示し、軸索輸送関連物質の蓄積様式から、レビー小体の形成が軸索輸送障害と平行して進行することを示した^<1)>。また、DLBはα-シヌクレインの蓄積により発症するシヌクレイノパシーとされているが、α-シヌクレインとリン酸化タウの免疫組織化学を用いて、DLBでは神経細胞のみならずグリア細胞にもリン酸化タウが蓄積し、有意に高い頻度でthorn-shaped astrocytes、coiled bodies、argyrophilic grainsなどのタウ陽性構造物が出現することから、タウオパシーの観点からDLBを捉える必要があることを示した^<2)>。さらに、パーキンソン病では脳幹のカテコラミン神経細胞がレビー小体を伴って脱落することが知られているが、チロシン・ヒドロキシラーゼ免疫組織化学を用いて、DLBでは脳幹のみならず大脳皮質と海馬のカテコラミン神経細胞が有意に強く脱落し、この中にはレビー小体を伴って変性するものがあることを示した^<3)>。本年度は、DLB以外にも、ユビキチンの蓄積する非定型ピック病(PiD)ではアセチルコリンのムスカリン受容体が減少すること^<4)>、タウオパシーである皮質基底核変性症(CBD)と進行性核上性麻痺(PSP)の病変が形態的にも生化学的にも共存し得ることを示した^<5)>。
