ヒト正常赤芽球系前駆細胞のサイトカイン惹起細胞内情報伝達系とその生理的意義の解明

研究概要

シグナル伝達実験には10^7個から10^8個レベルの均質な細胞が必要であり、正常なヒト造血前駆細胞を得ることが困難であったことから、これまでは主に株化細胞が研究に用いられてきた。造血細胞のシグナル伝達経路は検討に供する株化細胞によって異なり種特異性が存在する。本研究では、我々が開発したヒトCD34陽性細胞の大量純化法をもとに、赤芽球系前駆細胞(CFU-E)への系特異的分化・増殖誘導を行ない、これをモデルとして正常人の造血前駆細胞における、シグナル伝達機構の解析を進めるとともに、ヒト赤芽球系前駆細胞に発現する新規転写因子のクローニングを行なった。その結果、当該年度において以下の研究実績を得た。1)stem cell factor (SCF)は、Src family kinaseを介する情報伝達経路を介してヒト正常赤芽球前駆細胞のFas/Fasリガンド誘導アポトーシスを阻害する(Exp.Hematol 29:19, 2001)。2)SCFはPI-3K/AKT活性化経路を介してヒト正常赤芽球前駆細胞のアポトーシスを抑制する(Exp.Hematol 29:833, 2001)。3)Forkhead familyに属する転写因子FKHRL1がヒト正常赤芽球系前駆細胞およびヒト正常巨核球系前駆細胞の生存に関与する(Blood 96:941, 2000, J Biol Chem 276:15082, 2001)。4)ThrombopoietinはBc1-xLおよびPI-3K経路を介してヒト正常巨核球系前駆細胞のアポトーシスを抑制する(J Biol Chem, in press)。5)ヒト赤芽球系前駆細胞に発現する転写因子ZNF317の新規クローニングに成功しその機能を解析した(Biochem Biophysic Res Com. 288:771-779, 2001)。
