変異FLT3分子シグナルがもたらす細胞増殖・分化の脱制御機構の解析

研究概要

FLT3遺伝子の部分的なtandem duplication(FLT3/ITD)は、急性骨髄性白血病の約20%に認められる新たな予後不良因子である。本研究においては、FLT3分子の活性化におけるJM領域の役割を明らかにするために、JM領域のITD、欠失、点突然変異FLT3分子を作製し、その活性化機構を検討したところ、野生型JM領域はリガンド非存在下におけるFLT3分子の自己活性化能を抑制していることを示唆する結果を得、また、ITD変異JM領域は、キナーゼ領域が無くとも野生型FLT3分子と結合することにより恒常的活性化を誘導する知見を得た。一方、ITDや欠失変異体においては、その長さ、部位に関係なく二量体を形成し恒常的活性化をもたらしていることを明らかとした。更に、これらの複合体形成においては、分子シャペロンHSP90が介在分子として重要な役割をしていることを証明した。また、これら変異FLT3分子を32D細胞に発現させると、MAPK、STAT5、SHCなどのシグナル伝達分子の活性化が同様に認められた。一方、FLT3発現32D細胞は、G-CSFによる成熟好中球への分化が抑制されているが、PKCモジュレーターと分子シャペロン阻害剤の処理によって、G-CSFによる分化能が回復されることを見出した。更に、変異FLT3遺伝子トランスジェニックマウスの作製を施行中であるが、発現F1マウスは不妊傾向にあり、現在数種類の発現ベクターを用いることにより、発現量を調整し作製・樹立にむけ継続実験中である。
