ヒトT細胞白血病ウイルスI型TaxによるTGF-βシグナル伝達の抑制

研究概要	Transforming growth factor-β(TGF-β)は造血細胞を含む多くの細胞の増殖を抑制する増殖抑制因子である。TGF-βのシグナル伝達に異常が起こると細胞は自立性に増殖し、この結果、細胞のがん化につながる。TGF-βのシグナルは細胞表面のI型とII型の複合体よりなるセリン/スレオニン受容体によって受け取られ、Smadを介するシグナル伝達により核内に伝えられる。平成12年度の研究実績として、HTLV-1のトランスフォーミング蛋白Taxは転写共役因子であるp300/CBPをSmadと競合することにより、TGF-βシグナル伝達を抑制することを明らかにした。HTLV-1以外にも生体にがんを引き起こすウィルスが知られているが、平成13年度はHTLV-1と同じレトロウイルスであるHIV-1のTax蛋白やヘルペスウイルスであるEBウイルスのLMP-1蛋白もTat蛋白と同様にTGF-βシグナル伝達を抑制することを明らかにした。しかしながら、その分子機構はレトロウイルスとヘルペスウイルスで異なるという興味ある知見を得た。HIV-1のTat蛋白はTax蛋白と同様にp300/CBPと結合し、これら共役因子をSmadと競合することがTGf-βシグナル伝達抑制の分子機構であった。一方、LMP-1の場合は、その生物活性(NF-κB活性化など)を担う領域CTAR-1とCTAR-2がTGF-βシグナル伝達の抑制に重要であった。LMP-1自身はp300/CBPと結合できず、IκBαやIκBβ-の優性抑制変異体の導入により、LMP-1のTGF-βシグナル伝達の抑制は解除された。すなわち、LMP-1はNF-κB活性化を介して、TGF-βシグナル伝達を抑制することが明らかとなった。またCBP/p300発現プラスミドの導入は、LMP-1のTGF-βシグナル伝達の抑制を部分解除した。LMP-1はNF-κBを活性化し、NF-κBとSmadとの間でCBP/p300を競合することが、TGF-βシグナル抑制の分子機構である。以上の研究結果より、発がんに関与するウイルスの戦略として、TGF-βシグナル伝達の抑制はこれらウイルスの必須アイテムである可能性が示唆された。
