赤血球膜異常症における遺伝子発現制御とそのメチル化の意義

研究概要	ゲノムDNAの発現は、種々のpromoter昨日によって制御されていることが知られており、その一種にepigenetic controlがある。そこでgene methylationの立場から赤血球膜蛋白のうち、(1)β-spectrin(β-Sp),(2)band 3(AE-l),(3)protein4.2(P4.2)を対象に、各々の遺伝子であるSPTB, EPB3,ELB 42についてpromoter領域の5'-CpG-3' sitesのメチル化度を解析しその意義を検討した。方法はbisulfate genomic sequence法を用いた。 (1)SPTBおよびEPB3:SPTBでは、正常人35名(250クローン)の検討で、promoter領域に存在する5'-CpG-3'sitesは全て完全にunmethylated(メチル化度0%)であった。これに比べて、EPB3では、40名(588クローン)の検討で、全体として約2/3の5'-CpG-3' sitesに約80%以上の高度メチル化度を認めたが、一部にはメチル化度が約30%程度と低い5'-CpG3' sitesが存在した。この部位は、methylationに関して他の5'-CpG-3' sitesと異なる、特殊な機能部位であると推定された。 (2)ELB42:正常人23名(180クローン)の検索では、exon1のcoding region内の5'-CpG-3' sitesが約90%のメチル化度を呈するのに対して、promoter領域の5'-CpG-3' sitesは平均約66%のメチル化度であり中等度のメチル化度を示していた。続いてP4.2が赤芽球系細胞にのみ発現する蛋白である観点から、正常ヒト末梢血BFU-Eを用いた二相性液体培養法から得られた培養赤芽球を用いて同様に解析したが、これらの成熟段階にある赤芽球では、すでに末梢血単核細胞と同程度のELB42メチル化度を示していることが判明した。そこで、この培養赤芽球よりもさらに幼弱で均一な赤芽球系細胞群と考えられるヒト赤芽球系培養樹立株UT-7/Epoの細胞を用いて解析したところ、ELB42遺伝子promoter領域およびcoding regionの5'-CpG-3' sitesは全てunmethylated(メチル化度0%)であった。以上から、主要赤血球膜蛋白遺伝子の発現に関してmethylationの特異な態度が明らかとなった。
