白血病化に関与するMLL遺伝子の標的遺伝子の探索

研究概要

二次性白血病、乳児白血病の原因遺伝子であるMLL遺伝子は第11番染色体q23領域に存在し、t(4;11),t(9;11),t(11;19)などの転座により切断され、別の遺伝子とキメラタンパクを形成する。MLL遺伝子は転写制御因子であると考えられているがその標的遺伝子は明らかでなく、白血病化に関与する標的遺伝子を探索した。白血病細胞におけるMLL遺伝子転座を代表する変異MLLであるMLL-Zf(-)を安定発現する導入細胞(32D-Zf(-))と、ベクターのみの導入細胞(32D-VC)および正常MLLの導入細胞(32D-MLL)で、発現に差のある遺伝子をRDA(representational difference analysis)法で解析した。平成12年度は、32D-Zf(-)をtester、32D-VCをdriverとした解析を行ない、PI-3 kinase遺伝子が32D-Zf(-)で発現していることを同定していた。しかしながら、このPI-3 kinase遺伝子は正常MLLの導入細胞である32D-MLLでも発現が見られ、これのみでは白血病化の説明ができないことが判明した。
このため本年度は、32D-Zf(-)をtester、32D-MLLをdriverとしたRDA解析を行ない、32D-Zf(-)ではα-spectrin遺伝子が特異的に発現していることを見いだした。PI-3 kinaseを発現している32D-MLLへα-spectrin遺伝子を導入することにより、白血病化の特徴である細胞増殖促進と分化抑制が見られるかどうかは未確認であるが、MLL遺伝子転座によるPI-3 kinase遺伝子とα-spectrin遺伝子の発現誘導が白血病化に重要な役割を果たしていることが示唆された。
