細胞周期調整遺伝子による急性腎不全の遺伝子診断及び治療への基礎的検討

研究概要

平成14年度は、急性腎不全後の細胞増殖のkey factorとして、急性腎不全後の尿細管細胞の再生因子としてWnt4に注目した。Wnt4は、胎生期に腎に発現し、metanephric mesenchymeの分化に必須な遺伝子である。急性腎不全ラットにおいて、虚血、再灌流後6時間より、近位尿細管において発現が上昇することを報告した。Wnt-4の発現部位はAquaporin1の発現部位と一致しており、近位尿細管を考えられた。またWnt-4は、PCNA陽性細胞に発現しており、in vivoで、増殖している細胞に発現していると考えられた。Wnt4を尿細管細胞に強制発現させたところ細胞周期調整遺伝子であるcyclin D1,cyclin Aの転写活性と蛋白発現が上昇し、細胞増殖が認められた。以上のことからWnt4は急性腎不全の回復期の細胞増殖のkey factorの一つと考えられる(Terada et al. Journal of the American Society of nephrology, in press)。また腎細胞のapoptosisは、急性腎不全時に認められることが、報告されているが、尿細管細胞のapoptosisを制御する因子としてphosphatidylinositol 3-kinase-Aktを同定し、報告した(Terada et al Kidney Int)phosphatidylinositol 3-kinaseのノックアウトマウスでは、シスプラチン誘導の急性腎不全において、アポトーシスが、より起こり、予後が悪いことを見い出した。
