摂食調節ペプチドによる神経内分泌系の制御機構

研究概要

1.レプチンによるluteinizing hormone(LH)およびprolactin(PRL)のsurge促進作用には、脳内のnitric oxideが有意な役割を果たしていることを見い出した。
2.肥満ラットで認められる視床下部・下垂体・性腺系機能障害の少なくとも一部は・高レプチン血症に起因していることを見い出した。
3.レプチンによるgrowth hormone(GH)分泌促進作用への種々の神経ペプチドの関与について検討する目的から、ラット視床下部の弓状核-正中隆起領域を種々の濃度のレプチンでperfusionし、growth hormone-releasing factor(GRF),somatostatin(SRIF)およびneuropeptide Y(NPY)の分泌変動を観察した。その結果、レプチンによるGH分泌には、GRFの分泌冗進とSRIFの分泌低下の両者が関与しているが、予想に反してNPYの有意な関与はないことが示唆された。
4.近年その摂食調節作用が知られてきたleukemia inhibitory factor(LIF), ciliary neurotrophic factor(CNTF)およびprolactin-releasing peptide(PrRP)が、LHおよびPRLのsurgeに及ぼす影響について検討した。その結果、これら3種のペプチドはいずれも両ホルモンのsurgeに対して促進作用を有することを見い出した。
