増殖型アデノウイルスを用いた甲状腺癌遺伝子治療の基礎的研究

研究概要

甲状腺分化癌と未分化癌に特異的な増殖型アデノウイルスを作成し、その効果をin vitroとin vivoにて検討した。
1、分化癌に対しては、甲状腺特異的プロモーター(サイログロブリンプロモーター)でアデノウイルスE1A(アデノウイルス増殖に必須)が発現するウイルス(ただしE1B19Kはウイルスプロモーターで発現し、E1B55Kは発現しない)を作成した。このウイルスをサイログロブリンを発現する分化型甲状腺癌とサイログロブリンを発現しない他の癌細胞に感染させ、in vitroとin vivoにおいてサイログロブリン発現甲状腺癌特異的なE1A発現・ウイルス増殖・細胞変性効果を確認した。今後は、殺細胞遺伝子や腫瘍免疫との併用療法の可能性を探るため、自殺遺伝子発現アデノウイルスや、サイトカイン発現アデノウイルスとの併用効果を検討する。
2、未分化癌に対しては、この種の癌は高頻度に腫瘍抑制遺伝子p53に変異を持つので、p53反応性プロモーターでCre recombinaseが発現するウイルスと、2つのloxPで挟まれたアデノE1A遺伝子がウイルスプロモーターで発現されるウイルス(E1B19Kは他のウイルスプロモーターで発現するように組み込まれているが、E1B55Kは発現しない)とを作成した。これらのウイルスをp53遺伝子に変異を持つ甲状腺未分化癌細胞とp53が正常な癌細胞に共感染させ、in vitroにおいてp53遺伝子に変異を持つ甲状腺未分化癌細胞特異的なE1A発現・ウイルス増殖・細胞変性効果を認めた。しかし、in vivoでは、in vitroに比較して共感染の確率が低いため、p53正常細胞においてもかなりの影響が見られた。よって、次はこの2つのウイルスを1本化し、in vivoでの効果選択性及び細胞変性効果の増強を試みる。
