骨形成因子(BMP)による骨修復能力を付加した新しい人工関節の開発

研究概要	1.新しいBMPの人工担体の開発 BMPの活性を生体内で作用させるために必要な担体として、新しい生体分解性ポリマーであるポリ乳酸-パラジオキサノン-ポリエチレングリコール共重合体(PLA-DX-PEG)を開発した。このPLA-DX-PEGをBMPと混合し、マウスなどの小動物に埋植して骨形成能を検討したところ、これまで用いられていた担体(コラーゲンなど)より優れた担体であることを確認した。 2.BMP/PLA-DX-PEG/チタン複合体による骨欠損の修復このPLA-DX-PEGとBMPの混合体をチタンメッシュインプラントに付着させた。ウサギの上腕骨に1.5cmの骨欠損を作成してインプラントを埋植してワイヤー固定した。4週間で骨欠損は完全に修復し、インプラント内部にも骨新生を認めた。 3.BMP/PLA-DX-PEGと人工関節の合体このBMP/PLA-DX-PEG混合体を実験用人工骨頭の表面に付着させ、イヌの大腿骨近位に大きな骨欠損を伴う人工股関節置換術を行った。結果は、人工骨頭周囲に新しい骨を誘導し、母床骨に強固に結合すると共に大きな骨欠損を修復した。さらに実験用人工関節のソケットを作成しBMP/PLA-DX-PEG混合体を付着させ、イヌの骨欠損を伴う人工関節全置換術の実験を行う予定である。 4.BMPの骨形成を促進する薬剤の検討実際にBMPを臨床でヒトに用いる場合には、BMPの骨誘導能力を促進して効率よく骨形成を得る薬剤の開発が期待される。この目的のために、ペントキシフィリンおよびロリプランを実験動物に投与したところ、BMPによる骨形成が促進することを見出した。
