ヒト尿道のエンドセリン受容体の検討とエンドセリンの腹圧性尿失禁治療への可能性

研究概要

本年度は,主としてヒト尿道組織を用いて、エンドセリン受容体についてcompetitive reverse transcriptase polymerase chain reaction(RT-PCR)を行ない、尿道各部におけるエンドセリン受容体の各サブタイプの量ならびに比を調べた。
【方法】:Åsbergらの方法(Scand J Clin Lab Invest 58:299-306,1998)に準じてendothelin A(ET_A)とendothelinB(ET_B)受容体サブタイプのmRNA並びにpreproendothelin-1(ET-1)mRNAのprimerならびにcompetitiorを作成し、膀胱腫瘍に対する膀胱尿道全摘症例の尿道標本にたいしてcompetitive reverse transcriptase polymerase chain reaction(RT-PCR)をおこなった。尿道標本は外尿道口から膀胱頸部までそれぞれ数切片に分断し、それぞれの部位におけるエンドセリン受容体の半定量を行い、その差異を検討した。作成したprimerは下記の通りであり、PCR産物はET_A=378,ET_B=333,preproET-1=446である。またcompetitorはTAKARA Competitive DNA constructin kitを使用し、ET_A,ET_B,preproET-1に対してそれぞれ250R,200R,300Rのcompetitorを作成した。

【結果】:男性尿道を近位尿道から遠位尿道まで4分割して測定したが、各部位ともET_A,ET_B,preproET-1の割合はほぼ一定で3×10^5〜10^6competitor量(copy)であった。女性尿道は近位尿道から遠位尿道までの3分割した。いずれの尿道部位でもET_A,ET_B,preproET-1ともほぼ一定で3×10^5〜10^6competitor量(copy)であった。
【結語】:男女とも尿道各部位のエンドセリン受容体のサブタイプ(ET_AとET_B)並びにpreproET-1の量はいずれもほぼ同量であり、かつ、部位による差異は見られなかった。即ち、全尿道におけるエンドセリン受容体のサブタイプはAとBともほぼ同量存在すると思われる。
