ヒト尿道のエンドセリン受容体の検討とエンドセリンの腹圧性尿失禁治療への可能性

研究概要	昨年度は、主としてヒト尿道組織を用いて、エンドセリン受容体についてcompetitive reverse transcriptase polymerase chain reaction(RT-PCR)を行なった。本年度(平成13年度)は各サブタイプのin situ hybridization probeを作製し、ヒト尿道筋組織における局在の差を検討した。【方法】:Asbergらの方法(Scand J Clin Lab Invest 58 : 299-306, 1998)に準じてendothelin A(ET_A)とendothein B(ET_B)受容体サブタイプのmRNA並びにpreproendothelin-1(ET-1)mRNAのprimerとcompetitior を作成し、膀胱尿道全摘男女症例(膀胱腫瘍)の尿道標本に対して、competitive reverse transcriptase polymerase chain reaction (RT-PCR)をおこなった。尿道標本は尿道を近位尿道から遠位尿道まで3-4分割し、各部位におけるエンドセリン受容体の半定量を行い、その差異を検討した。作成したprimerは下記の通りである。 sense antisense PCR products ET_A : ctctgcgctcttagtgttga acgctgcttaagatgttc 378bp ET_B : aatggacagcagtagaaattg gaccaggcaaagacggttttgg 333bp preproET-1 : tctacttctgccacctggacat ctcggttgtgggtcacataa 446bp またcompetitorはTAKARA Competitive DNA constructin kitを使用し、ET_A, ET_B, preproET-1に対してそれぞれ288bp, 244bp, 342bpのcompetitorを作成した。なおin situ hybridization probeも上記のantisenseをAlexa Fluor 488でラベルし、型のごとく45℃で14時間hybridizationを行い共焦点レーザー顕微鏡にて局在を検討した。【結果】:各部位ともETA, ETB, preproET-1の割合はほぼ一定で3×10^5〜10^6 competitor量(copy)であった。in situ hybridizationでは尿道の平滑筋全層にET_A, ET_B, preproET-1が認められ、サブタイプによる局在は見られなかった。【結語】:ヒト尿道各部位のエンドセリン受容体のサブタイプ(ET_AとET_B)並びにpreproET-1の量はいずれもほぼ同量であり、かつ、部位による差異は見られなかった。即ち、全尿道におけるエンドセリン受容体のサブタイプはAとBともほぼ同じ部位にほぼ同量存在すると思われる。
