子宮内膜癌のエストロゲン依存性増殖におけるサイクリン発現の分子機構の解析

研究概要	子宮内膜腺上皮細胞および一部の子宮内膜癌はエストロゲン受容体(ER)を発現しエストロゲン依存性の増殖を呈するがその機序は不明である。一方近年細胞増殖にはサイクリンをはじめとする細胞周期調節因子が深く関与することが明らかになった。今回の研究では、エストロゲン(E2)による子宮内膜腺上皮細胞およびER陽性子宮内膜癌Ishikawa細胞の増殖機序を細胞周期調節因子の観点から解析することを目的とした。血清除去した培養子宮内膜腺上皮細胞にE2を添加し各種細胞周期調節因子の発現をWesternblottingにて観察したところ、サイクリンがD1→E→A→Bの順に発現されたことから、子宮内膜腺上皮のE2による増殖にはサイクリンD1の発現が重要であると考えられた。次にE2によるサイクリンD1発現の機序を解明するために、サイクリンD1プロモーターを利用したルシフェラーゼアッセイを施行したところ、腺上皮細胞ではサイクリンD1遺伝子上流に存在する初期転写調節因子であるAP-1蛋白が結合するAP-1結合配列がサイクリンD1の転写活性に重要であることが判明した。また正常内膜腺上皮細胞ではE2によりAP-1蛋白が誘導されること、加えて、ゲルシフトアッセイにより、サイクリンD1プロモーターのAP-1配列はE2刺激した正常内膜腺上皮細胞の核蛋白と結合することが判明し、正常内膜のE2による増殖においてはAP-1を介したサイクリンD1発現が極めて重要であることが判明した。一方、Ishikawa細胞でもE2によってサイクリンD1が発現することが認められたが、ルシフェラーゼアッセイによりその発現にはAP-1を介さない経路の存在が示唆され、正常内膜とは異なるE2依存性増殖機序が存在する可能性が示唆された。
