精子細胞膜上のGPIアンカー蛋白を用いた免疫学的避妊ワクチンの開発

研究概要

これまでの我々の研究から、免疫性不妊婦人が主として血中に保有する精子不動化抗体の対応抗原として、GPI(glycosylphosphatidyl-inositol)アンカー蛋白の一種であるCD52と相同の蛋白として知られる精子細胞膜蛋白SAGA-1(Sperm agglutinating antigen-1)の存在することが判明した。SAGA-1とは精子の凝集、および精子の不動化に強く関与する蛋白である。そこでこの蛋白に対する抗体、すなわち精子不動化抗体を保有する免疫性不妊婦人の血清が、果たしてin vitroで受精や胚発生に与える影響につき、マウスの体外受精系を用いて検討した。その結果、ヒト精子を不動化する抗体を含む血清の多くは、マウスの精子も不動化することが明らかになった。さらにヒト精子とマウス精子をともに不動化する患者血清は、マウスの体外受精を有意に阻害することも判明した。また抗体を含む血清を添加した培養液中で発生したマウス胚は、有意に高率なfragmentationを伴うことも明らかになった。他方、男性不妊症患者のうち抗精子抗体を有する者において、射出精子上に精子不動化抗体を結合する患者が存在することを研究者らは発見してきたが、これらの患者は精子無力症をほぼ必発で伴うことも明らかにした。これらの事実から、精子不動化抗体は精子の運動性・受精・および胚発生などの過程において、不妊症の発生と密接に関連することが明らかとなった。従ってその対応抗原として注目されるGPIアンカー蛋白を精製することにより、今後免疫学的避妊ワクチンの開発が可能であることが示唆された。
