耳石器及び半規管入力の前庭神経核ニューロンへの統合

研究概要

半規管神経並びに耳石器神経を選択的に刺激し、標的とするニューロンあるいは交感神経活動を除脳ネコから記録した。(1)半規管入力と耳石器神経からの単一前庭神経核ニューロンへの収束パタンを明らかにした。どのペアの実験でも収束は主に前庭脊髄路ニューロンに多く見られ、眼運動系に軸索投射するニューロンへの収束は少なかった。(2)球形嚢系の直線加速度増強のメカニズムとしてCross-striolar inhibitionが重要でCommissural inhibitionは存在するが弱い。一方卵形嚢系ではCommissural inhibitionが強力でCross-striolar inhibitionは存在するが弱いことを明らかにした。(3)半規管系で検出される回転感覚は対側視床で中継される。ところが耳石器系で検出される直線加速度感覚は同側視床で中継されることが明らかになった。(4)両耳石器神経活動は交感神経系に対し抑制と興奮効果があることを明らかにした。(5)胸鎖乳突筋運動ニューロンへの両耳石器入力を調べた。卵形嚢神経を刺激すると反対側胸鎖乳突筋運動ニューロンへ2シナプス性興奮が、同側胸鎖乳突筋運動ニューロンへは2シナプス性抑制性電位が発生した。球形嚢神経を刺激すると同側胸鎖乳突筋運動ニューロンへ2シナプス性抑制性電位が発生し、対側への結合は無かった。我々の結果は、広く耳石器系の臨床機能検査で行われているvestibular evoked myogenic potentials(VEMP)の神経機構を明らかにしたことになる。(6)球形嚢神経を微小電流刺激し両眼全ての外眼筋運動ニューロンから細胞内記録した。上直筋、下直筋及び上斜筋運動ニューロンの一部にのみ過分極あるいは脱分極電位が発生した。球形嚢が垂直直線加速度を検出することを考えると合目的的結合である。
