口腔の感覚と運動発現に関与する三叉神経ニューロンの活動調節機構の機能形態学的研究

研究概要

ラット咬筋を切断した後の三叉神経感覚核・運動核におけるnicotinamide adenine dinucleotide phosphate-diaphorase(NADPH-d)及び神経型一酸化窒素合成酵素(n-NOS)の発現様式をそれぞれ酵素組織化学法及び免疫組織化学法を用いて時間経過を追って調べた。NADPH-dは三叉神経中脳路核では切断側に8週にわたる、運動核では両側性に一時的な新たな発現を認めた。三叉神経吻側亜核及び尾側亜核では両側性の一時的な発現の増加を認めた。n-NOSの発現はNADPH-dの発現と同様な経過を示したが、1週間遅れた。
c-Fosは痛み刺激に反応して脊髄/延髄の後角の浅層部ニューロンの核内に多く発現する転写調節因子である。c-Fosは多くのペプチドの転写を調節すると考えられているが、痛みの感覚をいかに調節するか不明である。アジュバントをラット後肢に注射して痛覚過敏を起こした後に発現するc-Fos発現をアンチセンス法を用いてブロックし、熱疼痛受容感覚の変化をプランターテストを利用して調べた。c-fosのmRNAに対するアンチセンス(AS)オリゴデオキシヌクレオチド(ODN)を腰髄に注入し4時間後アジュバントをラット後肢に注射した。AS ODN注入例ではアジュバント注射3時間後の脊髄後角のc-Fos発現をミスマッチ(MM)ODN注入例に比べ有意に減少した。さらにAS ODN注入例ではMM ODN注入例に比べ有意に熱疼痛閾値を増加した。従ってアジュバント注射により誘導される脊髄後角のc-Fosは疼痛感覚を維持したり強めたりするような転写を促進すると考えられる。
さらに第1次(SI)及び第2次(SII)大脳皮質体性感覚領野を電気刺激し、延髄後角におけるc-Fosの発現様式を調べ、SIIの電気刺激が口腔領域の疼痛受容感覚を減少させることを明らかにした。
