BMP2による骨芽細胞および脂肪細胞分化の制御機構の解明

研究概要	1.未分化間葉系細胞の骨芽細胞および脂肪細胞分化におけるC/EBPβの役割初代マウス骨芽細胞において転写因子C/EBPβが発現していることが明らかとなった。さらに未分化間葉系細胞株C3H10T1/2細胞にBMP2を作用させると転写因子C/EBPβの発現が誘導された。そこでC/EBPβの骨芽細胞分化に対する役割を検討するためにC3H10T1/2細胞にC/EBPβを過剰発現させ、その効果を検討したところ、ALP活性およぴオステオカルシンの発現が誘導された。以上の結果から、C/EBPβは、脂肪細胞のみならず骨芽細胞分化誘導作用を有していることが明らかとなった。 2.未分化間葉系細胞の骨芽細胞分化におけるC/EBPβとCbfa1の相互作用未分化間葉系細胞に骨芽細胞分化の制御に深く関わるCbfa1をC/EBPβとともに過剰発現させると、骨芽細胞分化、オステオカルシンプロモーター活性が著明に促進された。免疫共沈降実験の結果、C/EBPβがCbfa1と結合することが見出された。さらにC/EBPβは、オステオカルシンプロモーターに直接結合することが明らかとなった。 3. C/EBPβアイソフォームLIPの骨芽細胞および脂肪細胞分化に対する相反作用 LIPをC3H10T1/2細胞に過剰発現させると、BMP2刺激あるいはPPARγの過剰発現により誘導される脂肪細胞誘導作用が著しく阻害された。一方、BMP2存在下あるいはCbfa1存在下でLIPを過剰発現すると骨芽細胞分化が促進された。興味あることにLIPはCbfa1およびオステオカルシンへの結合能を有していることが示された。以上の結果より、未分化問葉系細胞の分化方向の決定には、C/EBPβアイソフォームLIPが、重要な役割を演じていることが示唆された。
