ヒト口腔扁平上皮癌における血管新生に関わる諸因子の研究

研究概要	近年、癌の進展過程における異常増殖および転移能の獲得が血管新生に強く依存していることが明らかにされてきている。本研究は、血管新生蛋白としても注目されているが、口腔領域の腫瘍ではいまだ報告されていないId1,Id2,Id3についての発現分布や発現様式を免疫組織学的にあるいはWestern blot法やin situ hybridization法でヒト組織と培養細胞株を用いて検討することである。また、リンパ節転移といった臨床因子や予後との関係やVEGFとそのレセプターでありリンパ管新生にも関与するFlt-4についても検索を行うことである。 (1)Idタンパクの発現については免疫組織学的に83例の口腔扁平上皮癌と対照群に8例の正常口腔粘膜を検討した。口腔扁平上皮癌においてはId1(67.5%),Id2(55.4%),Id3(47.0%)でId1の発現頻度が最も高く、それぞれ分化度が低くなるにつれてその発現がより強度になる傾向が認められた。凍結組織標本並びに扁平上皮癌培養株を用いて行ったウェスタンブロツト法による解析においても各Id蛋白の発現を確認した。In situ hybridization法を用いて24例の口腔扁平上皮癌を検討すると、正常組織に比べて有意にその細胞質に強い発現を認めた。(Oncol.に投稿中) (2)VEGF, E-cadherin, Ki-67,Flt-4の免疫組織学的検討ではFlt-4のみに有意に頚部リンパ節転移との相関を認めた。また、腫瘍の発育形態や厚さなどの臨床病理学的因子も転移と相関していた。(J.
