消化管の神経系を介して作用発現するトリテルペンサポニンの探索と機能開発

研究概要

平成12年度は、漢薬地膚子の主サポニンmomordin Icおよび西洋トチノキ種子サポニンescin類の胃排出能抑制作用および小腸内輸送能亢進作用に関与する神経メディエーターついて検討した。いずれのサポニンも胃排出能抑制作用にはドーパミン(DA)がメディエーターとして関与しており、DA_2受容体刺激によってプロスタグランジン類(PGs)を遊離させ、最終的に胃排出能を抑制させることを明らかにした。一方、小腸内輸送能亢進作用にはDAは関与しておらず、セロトニン(5-HT)合成阻害剤および5-HT拮抗薬を用いた実験結果から、5-HTがメディエーターとして重要であることが判明した。さらにPG合成阻害剤や一酸化窒素合成阻害剤を併用した実験結果から、5-HT_2受容体刺激によって一酸化窒素(NO)またはPGsを遊離させ、最終的に小腸内輸送能を亢進させることが明らかになった。また,Escin類のMg^<2+>吸収促進作用には小腸内輸送能亢進作用と同様、少なくともNOが関与することを証明した。
機能性サポニンの探索研究として、フェヌグリーク(Trigonella foenum-graceum、種子)から6種の新規フロスタノール型ステロイドサポニンtrigoneoside類を単離し、化学的および物理化学的データによりそれらの化学構造を明らかにした。胃粘膜保護作用が見出されたフサザクラ(Euptelea polyandra)葉から6種のノルトリテルペンサポニンeupteleasaponin類を単離し、それらの化学構造を明らかにした。茶(Camellia sinensis var.assamica)種子および葉から新規アシル化ポリヒドロキシオレアネン型サポニンassamsaponin F,G,H,I,Jを単離し、その化学構造を明らかにするとともに、主要サポニンtheasaponin E1,E2に胃排出能抑制および小腸内輸送能亢進作用を見出した。エンドウ豆(Pisum sativum、種子)からは新規オレアナン型サポニンpisumsaponin I,II、沖縄産ニガウリ(Momordica charantia)から新規ククルビタン型トリテルペンサポニンgoyaglycosides-a,-b,-c,-d,-e,-f,-g,-hおよびオレアナン型トリテルペンサポニンgoyasaponin I,II,IIIを単離し、それらの化学構造を明らかにした。
