消化管の神経系を介して作用発現するトリテルペンサポニンの探索と機能開発

研究概要

ニガウリ(Momordica charantica)果実から,ククルビタン型サポニンgoyaglycoside a-hおよびオレアナン型サポニンgoyasaponin I-III,エンドウ(Pisum sativum)種子からオレアナン型サポニンpisumsaponin I、II、ツルムラサキ(Basella rubra)地上部から糖が酸化分解して生成したと考えられる珍しい置換基を有するオレアナン型サポニンbasellasaponin A-D,フサザクラ(Euptelea polyandra)葉からノルオレアナン型サポニンeupteleasaponin Vl_XII,マリーゴールド(Calendula officinalis)花からオレアナン型サポニンcalendasaponin A-D、インドニンジン(Withania somniferae)根からエルゴスタン型サポニンwithanoside I-VII,ゴツコラ(Centella asiatica)地上部からウルサンおよびオレアナン型サポニンcentellasaponon A-D,ツバキ(Camellia japonica)花からノルオレアナン型およびオレアナン型サポニンcamellioside A-Dおよび田七人参(Panax notoginseng)根からダンマラン型サポニンnotoginsenoside L-Nを単離し,それらの化学構造を明らかにした.
これらのサポニンに糖負荷ラットでの血糖値上昇の抑制作用,胃排出能抑制作用,エタノール誘発の胃粘膜保護作用,薬物耐性の抑制作用,アルドース還元酵素阻害作用,血小板凝集亢進作用および免疫増強作用の(アジュバント作用)あることを明らかにした.また,糖吸収抑制作用や胃排出能訳載作用など消化管への作用が認められたサポニンについては,脳内および末梢組織中のドーパミンやセロトニンなどの神経伝達物質の定量を試みるとともに,二次元NOESY, ROESYスペクトルの測定やX線結晶解析、力場計算によってサポニンのコンホメーンョンを解析した.
