研究概要

1.昨年度の研究成果を踏まえ、収縮機能実験・電気生理実験、遺伝子発現実験を行うことにより、プロスタサイクリン(IP)受容体刺激に伴う血管弛緩反応におけるcAMP-依存性およびcAMP-非依存性機構を更に踏み込んで解析した。
2.モルモット大動脈では、非常に高密度のマキシKチャネル(α-サブユニット)のmRNA発現を認めた。
3.モルモット大動脈の単離平滑筋細胞を用いたパッチクランプ実験から、IPアゴニストであるベラプロストがGTP依存性にマキシKチャネル電流を増加させることを明らかにした。GTP非水解性アナログであるGTPγSでも同様の結果を得たが、GTPはそれ自身ではマキシKチャネルをほとんど活性化しなかった。
4.マキシKチャネルのクローンチャネル(α-サブユニット)を発現させたHEK細胞およびカエル卵母細胞を用いてベラプロストの効果を検討したところ、チャネル活性化効果は全く認められなかった。
5.Gs蛋白に作用するコレラトキシンで摘出大動脈標本を6時間処置すると、緩徐な血管弛緩が引き起こされた。熱処理したコレラトキシンではこの作用は消失した。
6.コレラトキシンの作用はアデニル酸シクラーゼ阻害薬であるSQ22536処置によっても影響されなかった。
7.コレラトキシンの血管弛緩作用は、SQ22536処置下および非処置下でもイベリオトキシン(マキシKチャネルの選択的チャネルブロッカー)により有意に抑制された。
8.以上の結果は、Gs蛋白がマキシKチャネルを直接制御してこれを活性化することを示し、IPアゴニストによるcAMP-非依存性マキシKチャネル活性化機構の一部を説明するものである。
9.現在、パッチクランプ法によりGs蛋白のα-サブユニットの効果を検討するとともに、微小電極法により、マキシKチャネル活性化の静止膜電位に及ぼす影響を検討している段階である。
