脳梗塞モデルにおけるグリア細胞の修復および神経保護作用の研究

研究概要	本年度は、ラットを用いた中大脳動脈閉塞-再灌流モデルを作成し、経時的に、神経細胞およびグリア細胞がどのように変化するかについて、カイニン酸脳室内投与モデルと比較検討した。2時間の中大脳動脈閉塞により線条体の約半分が神経細胞死に陥っていた。その後、4、12時間、1、3、7日後では、線条体の全体、さらに大脳皮質側頭葉の広範囲に神経細胞死が引き起こされた。この時、神経細胞死に先立ち、微小管関連蛋白質-2(MAP2)は消失した。しかしながら、多量の神経細胞死が起きているコアの梗塞巣においても、パーキンソン病レビィー小体の主成分およびアルツハイマー病老人斑の非アミロイド性成分として知られているα-シヌクレインおよびチトクロームcは正常時と変わらず、死んだ神経細胞に残存していた。一方、ストレス耐性蛋白質ヘムオキシゲナーゼ-1(HO1)は修復可能なペヌンブラ領域のミクログリアおよびアストロサイトに発現したが、神経細胞には発現していなかった。また、3および7日後ではコアの梗塞巣にミクログリアが浸潤していた。In vitro実験系で、精製チトクロームcは過酸化水素により凝集された。さらにα-シヌクレインは過酸化水素およびチトクロームcによりその凝集が促進されたが、グリア細胞には発現しなかった。以上のことから、脳梗塞-再灌流により発生した酸化ストレスはチトクロームcおよびα-シヌクレインの凝集性を促進し、神経細胞特異的に障害を引き起こすこと、一方 HO1を発現したグリア細胞は修復され、さらに梗塞巣に浸潤して、神経細胞を修復しようとしていることが推定された。今後、その詳細なメカニズムを解析していく予定である。
