ヒト単球中の高比重リポ蛋白(HDL)結合蛋白の機能および構造の解明と測定法の開発

研究概要	本年度、我々はFACScan-フローサイトメトリー装置(Becton Dickinson社)を用いてフルオロイソチオシアネート(FITC)標識高比重リポ蛋白3(HDL3)をリガンドとしたヒト末梢血単球中のHDL結合蛋白の結合活性の測定系の開発と、ヒト単核球分画からHDL結合蛋白の精製を試みた。さらに、HDL結合蛋白のリガンドとして超遠心分離法およびDEAE-イオン交換により調製したpre β HDLおよび脱脂して得たアポHDLの結合性について検討した。フローサイトメトリーを用いた測定系において、ヒト単球の細胞膜表面をトリプシンにより処理をおこなったところHDLとの結合活性は減少したことからHDL結合部位はヒト単球の細胞膜表面にHDL結合蛋白が存在すると示唆された。リガンドとしてHDL粒子だけでなくアポHDLとも特異的に結合したことからHDL結合部位はHDLの脂質ではなく蛋白を認識していることが見い出された。また、ヒト単球はHDLの亜分画であるpre β HDL(脂質含量が少なく、細胞からコレステロールの引き抜きに関与すると報告されている)とも親和性を有することを認めた。比重法によりヒト末梢血から単核球分画を調製し、界面活性剤により単球膜を可溶化した後、DEAE-イオン交換、レクチンアフィニティおよびHDLアフィニティカラムクロマトグラフィーによりHDL結合蛋白の精製を行った。さらに、逆相高速液体クロマトグラフィ(RP-HPLC)により純度の高い蛋白を得た。調製した精製蛋白を抗原としてマウスに免疫し、特異抗体を作製中である。
