研究概要

Fas-receptorに始まる死のシグナルは、起始部の膜受容体の構造異常か細胞内シグナルカスケードの異常かに大別される。今年度は、(1)膜受容体Fasのmissense point mutationとシグナル障害(BBRC296:1251-1256・2002)と(2)細胞内シグナル系のkey-蛋白caspase-8のaberrant formの異常発現とシグナル異常(Int J Hematol76:50-54.2002)について研究し、その成果を前述のジャーナルに報告した。
(1)Fas-signaling研究のモデルとしているFas高発現ATLにおいて、cell line化した2例においてFas mutation異常を見いだしている。今年度は、その内の一例(RSO4)についてmutationとFasシグナル異常の機序について研究した。解析の結果、Fasの細胞外領域に相当するexon2 313塩基がA-G置換し、アミノ酸がhistigineからarginineへ代わっていた。この変化は、molecular modeling法の解析にてcytine-rich domain(CRD)52-58でwild typeと異なる構造へ変化していることが予想された。その結果、シグナル伝達に不可欠な重合化が障害されて、ついにはanti-apoptoticに傾く機序が考えられた。
(2)12年度に発見したcaspase-8 variant(exon8/9の間に136bpをinsertionしたaltemative splicing fromを異常にup-regulateしていること)について、このinsertionはintronの一部の挿入でframe shin機序でストップコドンを生じる為に酵素活性部分の大部分を欠くtruncated casp-8となり、decoy作用にてFas-mediated apotosisを負に制御していることが予想されることを明らかにして、Int J Hematoloに報告した。
