植物の光屈性に関わる光誘起生長抑制物質

研究概要	植物の光屈性発現機構は『影側の生長促進によるものなのか、それとも光側の生長抑制によるものなのか』という論争が長年続いている。報告者は、数年来、ダイコン、ヒマワリ、キャベツ、トウモロコシなどの光屈性の研究を行い、個々の植物でそれぞれ固有の光誘起生長抑制物質が関与していることを明らかにしてきた。平成12年度は、光屈性発現機構の更なる解明を目指して、トウモロコシを研究材料に限定し光刺激による加水分解酵素の活性化および光誘起生長抑制物質の遊離と光屈性の関係を明らかにすることを目的とした。トウモロコシの生長抑制物質MBOAおよびDIMBOAの生合成欠損変異株は光屈性を示さないが、MBOAをラノリンペーストとして幼葉鞘の片側組織表面に投与した場合は、暗条件下においてもMBOAを投与した側に屈曲する。この屈曲はMBOAの生長抑制作用によると考えるのが妥当である。そこで、生長抑制作用を示さない前駆体(配糖体)を用いて同様な実験を行い、光刺激による加水分解酵素の活性化により投与した前駆体が加水分解を受け生長抑制物質の遊離が起こり、幼葉鞘は前駆体を投与した側に屈曲するかどうか調べた。また、トウモロコシには数種類の加水分解酵素が存在しそれぞれ重要な働きを担っていると考えられているが、光刺激により特異的にDIMBOA-glcを加水分解する酵素の活性化だけが起こるのかどうか、それぞれの加水分解酵素の活性化度を調べた。さらに、加水分解酵素の阻害剤(カスタノスペルミン等)を投与した後に光刺激を与え幼葉鞘の光屈性の有無を調べた。しかし、実験条件等が一定せず現在再検討をしている。この研究が計画通りに進めば、これまでの学説を覆す結果が得られるものと確信している。
