細胞の接着と運動の新しい制御機構

研究概要

私共は、新しい細胞間接着機構であるNectin-Afadin系と、新しい細胞運動の制御機構であるFrabin-Cdc42系を見出している。Nectin-Afadin系は、免疫グロブリン様接着分子Nectinとそれをアクチン細胞骨格に連結させるF-アクチン結合蛋白質Afadinから構成され、Cadherin-Catenin系と密接に関連しながら上皮細胞のアドヘレンスジャンクション(AJ)とタイトジャンクション(TJ)の形成を制御している。FrabinはF-アクチン結合蛋白質であり、かつCdc42低分子量G蛋白質の特異的活性化因子(GEP)である。本年度の本研究において、Nectin-Afadin系とFrabin-Cdc42系の機能と作用機構を解析し、以下の成果を得た。
1.Nectin-Afadin系の機能と作用機構:上皮細胞において、Nectin-Afadin系がCadherin-Catenin系に加え、タイトジャンクションの構成分子であるZO-1やJAMも細胞間接着部位にリクルートすることを明らかにした。また、Nectin-Afadin系が、神経シナプスに局在し、Cadherin-Catenin系と協調してシナプスの形成を制御していることを見出した。さらに、Nectin-Afadin系が精巣におけるセルトリ細胞と精子細胞との異種細胞間の接着に関与し、精子細胞の形態形成に必須たることを明らかにした。
2.Frabin-Cdc42系による細胞運動の制御機構:Frabinが、特定のF-アクチン結合タンパク質を認識し、直接的にアクチン細胞骨格に作用するとともに、その近傍でCdc42を活性化することにより、細胞運動を制御することを明らかにした。
このように、本年度の研究は予想以上に進展し、当初の目的をほぼ達成することができた。
