GFP変異体を用いた蛋白分子2量体形成の可視化と細胞内情報伝達研究への応用

研究概要

細胞の増殖・分化を担っているシグナル伝達因子の活性化機構は未だ不明な点も多いが、蛋白質の2量体化やリン酸化による蛋白質の修飾が活性化に重要な役割を果たしているらしい。そこで、蛋白質の2量体化を人為的に起こし、その活性化への効果を調べる。低分子量G蛋白質Rasの下流で相互作用するRaf-1に着目してRaf-1の2量体化がRaf-1の活性化に必須であるのかどうか?また、実際Raf-1が活性化される場所はどこか?また、どの位の分子数が活性化されると細胞の形態変化が導かれるのかを生物物理学と分子生物学的手法を用いて解明したい。2量体化の確認は生物蛍光蛋白質間で生じると考えられるFRETの技術を利用することにした。
今年度は(1)DNA gyrase BのN-末側サブドメイン(GyrB)と生物蛍光蛋白質(CFP,GFP,YFP,DsRed)との融合蛋白質を約95%の純度の組換え体として得、溶液系でCFPとYFPの2量体化に伴うFRETがCoumermycin添加により観察された。また、それら蛍光蛋白質が目的のイメージングに適しているかについて顕微鏡の開発も行った。役者の数を知るために重要であると思われるこれら生物蛍光蛋白質の1分子レベルでのイメージング並びに蛍光特性を調べ、次の様な結果を得た。(2)GFP,YFP,DsRedは1分子レベルでイメージングする事が出来、さらに1分子レベルでのスペクトル測定の結果より、GFPやDsRedのスペクトル重心のばらつきは従来1分子レベルでのFRETに用いられてきた蛍光色素(Cy3等)と同程度であり、1分子レベルのFRETに用いることが可能であることが解った。
現在、溶液系での結果をふまえ、Raf-1にGyrB-CFP又はGyrB-YFPを結合させた融合蛋白質を細胞内で発現させ、生きた細胞内でのRaf-1の2量体化を行い、2量体化と活性化との関係を実時間で観察する系の立ち上げを行っている。
