感覚入力制御のニューロン機構と分子機構に関する実験形態学的研究

研究概要

1.ラットの三叉神経脊髄路核尾側亜核において、P物質受容体(NK1)を介する一次神経情報の伝達が、少なくとも部分的にはnon-synapticに行われることを示唆する形態学的知見を得た。
2.ラットの三叉神経脊髄路核尾側亜核において、三叉神経核視床路ニューロンにおけるカルシウム結合蛋白質の共存関係を形態学的手法により検討した。
3.ラット三叉神経中脳路ニューロンについて、以下の事項を確証した。
(1)セロトニン作動性入力線維を縫線核群から受ける。
(2)GABA作動性シナプス終末を受ける。
(3)機能的なGABA_A受容体は発現するが、機能的なGABA_B受容体は発現しない。
4.ラットの三叉神経脊髄路核尾側亜核の浅層において、P物質含有軸索がGABA作動性ニューロンおよびグリシン作動性ニューロンにシナプスすることを形態学的に証明した。
5.ラットの三叉神経脊髄路核尾側亜核のスライス標本について、細胞内記録法と細胞内染色法を併用して、内在性ニューロンの形態と電気的性質を調べ、それに基づいて、4種類の内在性ニューロンを見出した。また、1層の投射ニューロンの形態と電気的性質を調べた。
6.ラット背側縫線核ニューロンのなかに、軸索側枝によって、線条体と大縫線核・巨大細胞網様核α部に同時に投射するものがあることを発見した。また、スライス標本を用いて、背側縫線核ニューロンのセロトニン免疫活性、形態、電気的性質を調べた。
7.ラット三叉神経脊髄路核尾側亜核の侵害受容関連ニューロンの多くがprotein kinase C-gammaを発現することを示唆する所見を得た。
