新規神経栄養因子としてのHGFファミリーの発現・機能解析と神経疾患への適用

研究概要

HGFおよびそのファミリー分子(HLP)の神経系における機能解析
1.大脳皮質層構造形成過程における機能:HGFと受容体(c-Met)が大脳皮質発生過程で細胞内局在も含めダイナミックに発現制御をうけた。初代培養大脳皮質ニューロンとスライス培養でのHGFの機能解析結果、HGFは細胞移動・神経突起伸長・神径細胞生存の複数ステップで重要な機能を果たしていると示唆された。2.筋萎縮性側索硬化症:変性疾患の中でも特に重篤で、治療が困難である筋萎縮性側索硬化症(ALS)に対するHGFの治療効果を検討した。まずALS発症モデルマウス(ALS-Tgマウス)では疾患の進行に伴い、脊髄組織におけるHGFとそのレセプター(c-Met)の発現が促進されることを見い出した。そこで神経特異的にHGFを発現するトランスジェニックマウス(NS-HGF-Tgマウス)を作成し、このマウスをALS-Tgマウスと交配させることにより、HGFのALS発症抑制効果を調べた。その結果、NS-HGF/ALS-TgマウスではALS-Tgマウスに比べて運動神経の軸索変性および細胞死が著明に改善されており、それに伴いALS発症時期の遅延ならびに寿命の延長が認められた。さらにHGFによるALS進行遅延効果は運動神径への直接作用に加えて、グリア細胞の特異蛋白質(グルタミン酸トランスポーター等)を介することを明らかとした。3.Gas6の新しい神経栄養因子としての同定:脳神経系特異的チロシンキナーゼ型受容体"SKY"のリガンドGas6が神経栄養作用をもつことを明らかとした。4.脳神経系に発現する新規スカベンジャー受容体(SRCL)のクローニング:脳神経系においてスカベンジャー受容体は、ミクログリア等に発現し重要な機能をはたしている。本研究で、細胞外にC-typeレクチンドメインをもつ新しいスカベンジャー受容体をクローニングした。
