グリア芽細胞の移動メカニズムの解明:二種類の螢光分子で標識された生きた細胞の観察

研究概要

中枢神経系の発生過程における神経芽細胞・グリア芽細胞の移動メカニズムを知る目的から、これら未分化細胞の移動・運動動態を生きた状態で長時間観察した。
新生児ラット前脳のSVZから産出した未分化細胞の一部は、脳梁を通って対側大脳半球に移動することを見出した。その際、このレトロウィルス標識細胞は未分化な形態を保ちつつ、単一の先導突起を線維束に平行に伸しながら移動した。新生児期では、ラジアルファイバーは脳室壁から垂直に脳表に向かって伸びており、脳梁を通って伸びているものは観察されなかった。即ち、脳梁における細胞移動はラジァルファイバーの伸長方向とは直交する(tangentialな)方向を取ることが解った。一方、脳梁は燐酸化ニューロフィラメントで標識され、また電顕的には未だ髄鞘で巻かれていない未熟な軸索からなっていることを確認した。新生児期にレトロウィルスを注入された個体を成熟期に達した時点で固定し、標識細胞の最終分化像とその分布を観察すると、注入を受けた側と半対側に認められた標識細胞の多くは半球内側の白質・皮質に分布し、これは脳梁線維の投射部位におよそ一致していた。対側半球に分布していた標識細胞の数は注入側半球内のそれの1%程度であり、これらは分化したアストロサイトおよびオリゴデンドロサイトであり、単独あるいは2〜3個のclusterを形成していた。このように、SVZで産出したグリア芽細胞の部は脳梁を通って対側大脳半球に移動することが示された。このことは、両半球間でクローンの混合が起る可能性を示唆している。このtangentialな移動は、ラジアルファイバーではなく、軸索に沿って起こるものと考えられた。
