細胞呼び寄せ型人工血管用弾性マトリックスの設計に関する基礎研究

研究概要

本研究は平成12年度〜14年度の3年間の研究計画によって構成されている。平成12年度の計画は弾性線維のコア蛋白質エラスチンの高ホモロジー領域に共通して存在して弾性機能を発現するペンタペプチド配列の繰り返しポリマーである(Gly-Val-Gly-Val-Pro)nを基盤にして、フィブロネクチン、ラミニン等の接着蛋白質に共通して存在する細胞接着性ペプチド配列Gly-Arg-Asp-Ser-Pro、抗血栓性ペプヂド配列Gly-Arg-Asp-Tyr(Me)-Pro,及び細胞呼び寄せペプチド配列Val-Gly-Val-Ala-Pro-Glyをブレンドしたブロック型ポリペプチドを合成し、そのブレンドの最適化を検討した後、γ線照射して架橋させた細胞接着性・抗血栓性の細胞呼び寄せ型弾性マトリックスを作製することである。しかし、合成したGly-Arg-Asp-Ser-Pro配列が細胞接着性以外に血小板凝集阻害作用を有することを見出し、その作用機構を検討した結果、Gly-Arg-Asp-Ser-Pro配列は血小板上のgpIIb/IIIaに結合してフィブリノーゲンが血小板に結合することを阻害することにより、血小板凝集を抑制することが明らかになったので、Gly-Arg-Asp-Tyr(Me)-Pro配列の導入を中止して、代替としてGly-Arg-Asp-Ser-Pro配列の導入に変更した。現在基盤となる弾性ポリペプチド(Gly-Val-Gly-Val-Pro)n、細胞接着性及び抗血栓性ペプチドGly-Arg-Asp-Ser-Pro、及び細胞呼び寄せペプチドVal-Gly-Val-Ala-Pro-Glyの作製は完了したので、今後、最適化を検討し、架橋重合体の弾性マトリックスを作製する予定である。
