植物組織の形態形成における小胞輸送のダイナミクスと機能に関する研究

研究概要

高等植物の液胞の動態を生きたまま観察するために、液胞の水チャネルγ-TIPとGFPとの融合タンパク質を発現する形質転換シロイヌナズナを用い、共焦点レーザー顕微鏡で観察した。しばしば液胞内に強い蛍光を発する球状やチューブ状の構造が形成されているのを見出した。この構造("bulb")は、直径が1-3μmで、外側の液胞膜と連続していた。非形質転換体でもbulbが存在することを確かめるために、野生型の子葉を金属圧着型の急速凍結で固定して透過電子顕微鏡観察した。その結果、bulbはやはり存在しており、人工産物でないことが示された。連続切片像からの立体構築により、bulbは2枚の液胞膜に挟まれた細胞質が、液胞内に陥入するような複雑な構造であることが解った。一方、他の液胞マーカー、AtRab75とGFPとの融合タンパク質を発現させても、bulb様の構造は全く観察されなかった。しかし電子顕微鏡観察するとbulbは存在していた。つまり、構造は存在するが、GFP-AtRab75分子はそこから排除されていることを示す。また、γ-TIP-GFPにより可視化されたbulbにおいて、GFPマーカーの蛍光強度を測定したところ、外側の液胞膜に対して約3倍の強さであることがわかった。このことは、bulbの領域でγ-TIP-GFP分子が濃縮されていることを示す。
以上の結果から、新たに見出された液胞膜上の構造bulbは、連続した液胞膜の一部で複雑な立体構造を取り、特定のタンパク質が選別されているサブ領域であることを提唱した。
