研究概要

ウイルスが宿主に感染した際には、細胞内で様々な分子応答機構が起こる。本研究は、カイコ核多角体病ウイルス(BmNPV)と宿主細胞のBmNを材料としてBmNPVがBmN細胞に感染した際に起こる遺伝子発現プロファィルの変化の解析を行うことで、ウイルス感染時における分子応答を網羅的に解析することを目的とする。そのために、ウイルス宿主域決定メカニズムを解析するためのDNAチップ技術の導入と、ヒストンアセチル化によるヌクレオソーム構造の構造変化とバキュロウイルス遺伝子発現の関連について解析を行った
約20個のウイルス遺伝子に対する合成オリゴをスポットしたDNAチップを用いて、AcNPVを宿主Sf9細胞、2種類の非感染細胞BmN、S2へ感染させた際のウイルス遺伝子発現カスケードの差異を検討した。Sf細胞では感染5時間で4種類の遺伝子が未感染に対して転写量が増大していた。5時間から8時間にかけては半数以上の遺伝子転写量が増大していた。しかしBmN、S2細胞では感染時間経過に対して転写量が増大している遺伝子は少なく、特にほとんど感染が成立しないとされているS2細胞では有意に転写量が増大している遺伝子はなかった。
またヒストンアセチル化とウイルス遺伝子発現に関しては、アセチル化ヒストンH3抗体を使ったChromosome immunoprecipitationを各ステージの感染培養細胞に対して行った。ie-1プロモーター領域はPCRにより増幅された。さらに発現ステージが違うほかのプロモーター領域に関してもPCRによる増幅が確認されたので、アセチル化ヒストンはウイルスゲノムが核内に進入後、ゲノム全体に対してヌクレオソーム構造をとり、転写に関連することが推察された。
