ラット精巣におけるSKD3の機能解析

研究概要

近年、哺乳類精細胞分化における熱ショック蛋白質(HSP)の重要性がクローズアップされ、新たなメンバーがクローニングされるようになった。しかし、その機能のみならず局在すら明らかになっていないものが多い。本研究では、新たにクローニングしたHSPファミリーのひとつであるラットSKD3を用いて、そのmRNAおよびアミノ酸構造及び精巣における局在といった基礎データを収集するとともに、機能の全貌を明らかにし、精子発生の分子機構を明らかにすることを目的とした。
予備実験で行ったmRNAの精巣における局在および作製した抗体を用いたSKD3蛋白質の局在、mRNAの組織特異的発現パターン、腫瘍株化ライディッヒ細胞(LC540)におけるSKD3蛋白質の局在について再調査し論文投稿した(投稿中)。次に、真核細胞発現ベクターにSKD3遺伝子を組み込み、LC540細胞に導入した(電気穿孔法)。導入したLC540細胞におけるSKD3蛋白質の変動をwestern blottingにより解析したところ、発現量は想定されるよりも少なかった。現在、発現ベクターを数種類選定し、発現量が大幅に上昇するベクターを検索中である。今回のSKD3遺伝子を導入したLC540細胞の形態を光学顕微鏡および電子顕微鏡を用いて観察した。結果、両者の解像レベルで、遺伝子を導入していないLC540細胞との有意な違いは認められなかった。今後、SKD3蛋白質を多量に発現させたLC540細胞を用いて、その形態変化を詳細に検討する予定である。来年度には同LC540細胞を用いて、細胞内の各種ステロイド代謝酵素の変動を検討する予定である。
