鳥類松果体細胞における光受容および計時機構の解析

研究概要	生物は地球の自転や公転が作り出す環境の周期的変化に適応すべく自己体内に計時機構(生物時計)を具備したと思われる。鳥類ではこの計時機構は松果体に局在していることがすでに知られているが、松果体の時計のリズムがどのようにして行動や体温などのリズムに反映されているのかは、未だ謎である。本研究目的は、この時計と生理機能のリンクの仕組みを明らかにすること、加えて、松果体から分泌されるメラトニンはどのような機構で合成が制御されているのかの一端を明らかにすることにある。著者は、これまでの研究において、鳥の松果体には時計機構、光受容機構、およびメラトニン分泌機構が一個一個の細胞に存在し、お互いにリンクしていることをすでに示した。このことから考えれば、時計のリズムはメラトニンを介して生理機能のリズムを駆動している可能性が高い。そこでメラトニンをスズメやウズラに単一投与し、体温や行動におよぼす効果を検討したところ、投与直後から、急激な体温低下と行動抑制が認められた。この事実と、夜間にメラトニンの旺盛な分泌が起こることを併せて推測すれば、夜間、これらの鳥類は内因性メラトニンにより体温低下、行動抑制、睡眠が誘起されているものと考えられ、夜に昼行性鳥類が動けなくなることを良く説明できるように思われる。次に、ラットの松果体においてメラトニン合成を促進することが最近報告されているpituitary adenylate cyclase activating polypeptide(PACAP)のヒヨコ松果体細胞におけるメラトニン合成への関与の有無を検討した。その結果、PACAPは量依存的にメラトニン合成を促進し、その作用はvasoactive intestinal polypeptide(VIP)のレセプター拮抗剤で阻止されなかった。またPACAPに特異的なレセプターmRNAの発現をRT-PCR法で確認した。以上の結果は、ヒヨコ松果体細胞膜上にはPACAPに特異的なレセプターが存在し、メラトニン合成系はこのPACAP系によって一部制御されていることを示唆するものである。
