牛疫ウイルス膜蛋白の病原性発現機構における役割

研究概要

本研究の目的は牛疫ウイルス(RPV)のウサギ馴化株であるRPV-L株の膜蛋白Hの病原性発現への関与の有無と細胞親和性に必要な部位を検索することである。RPVの実際の宿主であるウシを用いた感染実験では世界的にも数少ない特殊施設でしか行うことができないが、RPV-L株はウサギでの感染実験系において牛疫の病態を忠実に再現する優れたウイルス株で、これまでに我々は感染ウサギ体内での病態の詳細な解析を行ってきた。本年度はまずRPV-L株のウイルスクローニングを行い最終的に10クローンを得た。それぞれについてウサギの感染実験を行った結果、1クローンを除き4日目をピークとして体重は100〜200g減少し、体温は40℃を越え、白血球数は約2000/μlまで減少した。一部は4日目までに死亡し、残りは体重・体温・白血球数ともに4日目以降回復傾向を示し、クローニング前のRPV-L株と同様の症状を再現することが確認された。それらの中で特に2つのクローンは感染ウサギ3例とも3日目に死亡し、再現性が高く病原性が強いウイルスクローンであることが明らかになった。一方、別な1クローンは病原性を示さない弱毒株であることがわかった。それぞれのクローンについて現在遺伝子解析を行っており、強毒株では全長の70%以上にあたる塩基配列を決定した。またH遺伝子については、ウシでのワクチン株でウサギに対して病原性を示さないRPV-RBOK株のH遺伝子と入れ換えた組換えRPV-RBOK(Lap-H)の作製に成功した。
