ウイルスベクターによる樹状細胞への高効率遺伝子導入と癌免疫遺伝子治療への応用

研究概要	平成12年度は主にヒト樹状細胞への遺伝子導入法の確立を目指した研究を行った。ヒト樹状細胞には末梢血単球よりGM-CSFとIL-4を用いて誘導した樹状細胞を使用した。最初にレトロウイルスベクターMFG-EGFPを用いてamphotropic packaging cellであるCRIP細胞からhigh titerのレトロウイルス液を作成し、我々の開発した遠心法を用いて遺伝子導入を試みた。しかしながら、フローサイトメトリーにて検討した遺伝子導入効率は5%以下で、末梢血単球由来樹状細胞へのレトロウイルスシステムを用いた遺伝子導入は困難と思われた。そこでアデノウイルスベクターAxCAEGFPを作成し遺伝子導入効率を検討した。通常の感染方法では、アデノウイルスを用いた場合においても高い効率で遺伝子導入するには高濃度のウイルスが必要であった。一方、アデノウイルス感染時に37℃で2000xg、2時間の遠心を併用することで10〜50MOIで遺伝子導入効率は85%以上に向上すると共に、遺伝子発現強度も有意に増加した。また、本法で遺伝子導入した樹状細胞の生存率は遺伝子導入をしていない樹状細胞のそれと同等で、混合リンパ球反応におけるリンパ球増殖刺激効果も保たれており、樹状細胞へのダメージは認められなかった。種々の細胞表面抗原の発現にも有意差は認められなかった。そこで実際に樹状細胞機能を増強する可能性のある遺伝子の導入が可能かどうか検討した。マウスモデルにおいてインターロイキン12(IL-12)の遺伝子導入により抗腫瘍効果が増強するという我々のこれまでの検討をもとに、ヒトIL-12を発現するアデノウイルスベクターを用いて遺伝子導入を試みた。その結果、遠心法を併用した場合においてIL-12遺伝子導入樹状細胞からのIL-12p70蛋白の産生が認められた。平成13年度はヒトIL-12遺伝子導入樹状細胞の機能解析を行うと共にヒト腫瘍抗原遺伝子導入樹状細胞の作成とその効果について検討する予定である。
