研究概要	海産の無脊椎動物であるホヤは、ある種の血球(バナドサイト)の液胞内にバナジウムを高濃度に濃縮している。パナドサイトの液胞はきわめて低いpHに維持されており、バナジウムを高濃度に濃縮している種ほど液胞内のプロトン濃度が高いという関係があることから、バナジウムの濃縮にプロトンの電気化学的エネルギーが共役している可能性が強く示唆される。本研究は、このエネルギー共役系を明らかにするための第一ステップとして、プロトンの濃縮に働く液胞型プロトンATPase(V-ATPase)の機能解明を行うことを目的として、(1)ホヤのV-ATPaseサブユニットをコードする遺伝子のcDNAを単離した後、(2)得られたcDNAを酵母突然変異体に組み込んで、機能解析を行う。本研究によって、プロトン輸送に携わるホヤのV-ATPaseの高機能のサブユニットの特定をめざすとともに、低pHの液胞を持つ酵母変異体を得ることを目指す。 1.スジキレボヤAscidia sydneiensis sameaの血球を採取しmRNAを抽出した。 2.データベース検索によって他種生物のV-ATPaseのサブユニットE〜Hおよびa, cの保存されている領域に対応するPCRプライマーを設計し、PCRを行った。 3.ホヤのV-ATPaseのサブユニットCの遺伝子により、酵母突然変異体を形質転換した。 4.形質転換した酵母の細胞内で確かにホヤ由来の遺伝子が転写されていることを確認した。 5.形質転換した酵母が、中性の培地上で増殖できない表現型を回復することを見いだした。
