カルシウムに対する高親和性・新規EF―hand蛋白質の三次元構造解析

研究概要	平成12年度はΔN32CBP40のカルシウム非存在下での構造解析を分解能3.0Aで解析し、構造精密化により三次元構造を明らかにすることができた。13年度はカルシウム存在下での結晶化を幾度か試みたが結晶化は困難であった。そこでカルシウムをソーキング(浸透)して得られた回折強度データを用いて、差フーリエ法によりカルシウム結合状態の構造解析を行った。カルシウム非結合の構造とカルシウム結合の構造の変化はカルモジュリンの様に大きくはなかった。一つは結晶内分子間パッキングの影響で、構造変化が抑制されてしまった可能性があるが、ΔN32CBP40は、EF1/2とEF3/4の間にカルモジュリンのようなリンカーがないため、もともと両者が相互作用してコンパクトな構造になっているので、カルシウムを結合しても大きな構造変化が起こらない可能性がある。このΔN32CBP40が非常に高いカルシウムに対する親和性を持っているのは、カルシウムN末端のαヘリックスからなるcoiiled-coilドメインと球状のEF-handドメインの部分がは広範囲にわたって分子内相互作用することによるものであることも構造からわかった。さらに、カルシウムによる自己集合に関しても、確かにカルシウムを結合したときの構造変化は少ないものの、ΔN32CBP40のN末端のcoiled-coilでは、1本目のヘリックスが2本目に比べて短い。しかし、二次構造予測からは、32残基が加わると2本のヘリックスの長さがそろい、coiled-coilが安定化・長くなると予想される。そのメカニズムはCBP40全長にカルシウムが結合すると、EF-handの構造変化がドメイン間相互作用を介してcoiled-coilに伝わり、coiled-coilのヘリックス間の相対配置が変化し、それによって、分子間でcoiled-coil同士がバンドルを作って会合するのではないだろうかと考えられる。本研究で得られたデータは現在Nature Structural Biologyに投稿中である。また、更にΔN32CBP40とCaMのキメラについても結晶を得ることができ、現在構造解析中である。
