駿河湾深層水利用に関する低次生物生産構造の解明と有機物の特性把握

研究概要

1.閉鎖系の培養実験による化学的因子と生物的因子の相互把握
表層水、397m、687mの深層水各60lを滅菌済みのポリカーボネイトボトルに採取し、屋外に設置した水糟で培養を行った。水温は約25℃、光量は自然光の約10%とした。サンプリングは1日おきに11日間行い、1回おきに対照として同条件で遮光したサンプル(397m,687m)についてもサンプリングを行った。栄養塩、POC/N、DOC/N、植物および動物プランクトンの種組成、バクテリア、シアノバクテリアおよびNano-flagellate数、クロロフィル-aおよびカロテノイド類、基礎生産量を測定した。POC(粒子状有機炭素)の初期濃度は表層水、397m,687mでそれぞれ177,31,33μgC/lであった。表層水ではPOCは2日目に419μgC/lと最大になったのに対し397m、687mではそれぞれ1877μgC/l(9日目)3186μgC/l(11日目)となった。植物プランクトンの存在量は、表層では初日2.3×10^4から最大5.1×10^5となり、深層水では初日に20-150cells/lであったのが、2.7×10^6cells/l(397m)、6.6×10^6cells/l(687m)まで増加した。深層水中の優先種は、Thalassiosira sp(f.minima) Chaetoceros costatum, Skeletonema costatumであり、サイズは1〜5μmがほとんどであった。パクテリアやnano-flagellateは7〜9日月に顕著な増加傾向を示した。
2.駿河湾深層水の有機物特性の把握
POMの組成は深層水ではたんぱく質が40%、核酸が5%、脂質は23%、炭水化物は13%、未知同定が19%である。DOMの組成は深層水でアミノ酸は5%以下、炭水化物は10%、脂質は8%で、残りの80%近くは未同定である。植物プランクトンによる基礎生産量として駿河湾では1.4μgCl^<-1>day^<-1>(Dec.2001)から13.2μgCl^<-1>day^<-1>(July,2001)の範囲にある。駿河湾深層水の蛍光スペクトルを測定したところ標準品である「腐植物質」と極めて類似のスペクトルが得られた。酢酸エチルで抽出された化合物の^1H-NMRの解析から、グリセリドが主成分であることを明らかにした。
