研究概要

内分泌攪乱物質の生殖細胞に及ぼす影響を半数体精子細胞特異的遺伝子群の発現レベル(短期的影響)及び精子形成障害(長期的影響)を指標として明らかにすることを目的として研究を行った。今年度は長期的影響について検討するために、エストロゲン様構造を持つBisphenol A(BPA)の妊娠雌個体に対する微量暴露による胎児への影響を出生後雄マウスの生殖器官について検討した。4週齢雌マウスにBPAを2ng/kg体重/day週3回経口投与し、投与5週目に交配、分娩前日まで投与を継続した。処理雌マウスから生まれた新生児の体重、雄の妊よう性、生殖器官の重要、精巣の組織学的観察を行った。
得られた結果
1.処理群新生児の体重増加率を非処理群のそれと比較すると離乳期に至るまでは増加率が低く、離乳後は回復した。出生時では体重差が見られないことから、母体に対するBPAの影響が授乳に現れる可能性が示唆された。
2.出生後7週齢の雄において精嚢および精巣重量の低い個体が見られたが、14週齢では差が見られず、妊よう性にも異常はなかった。このことは幼弱期に現れる影響が成体になり回復する可能性を示す。
3.精巣の組織学的検索では14週齢で生殖細胞の減少およびセルトリ細胞からの剥落等の異常所見が認められる個体が存在した。このように精巣重量、妊よう性等には著しい障害は認められないものの、精子形成に対しては明らかな影響が認められた。
BPAは生活環境において多用される合成樹脂であるポリカーボネートの原材料として使用されている。BPAが、体重kg当たりngという極微量の経口投与を受けた母親から母子移行により胎児に作用し、その影響が生後14週の個体の精巣に現れることが本研究において示された。この結果は生活環境中の化学物質による生殖への重大な影響であり、今後生殖細胞で発現する遺伝子群に対する影響を含め、分子レベルでの作用について詳細に検討する予定である。
