研究概要

内分泌撹乱物質の生殖細胞に及ぼす影響を半数体精子細胞特異的遺伝子群の発現レベル(短期的影響)及び精子形成障害(長期的影響)を指標として明らかにすることを目的として研究を行った。食品容器に多用されるプラスチック製品のポリカーボネイトの原料であるBisphenol Aにエストロジェン作用のあることが判明していることから、本研究においてはBisphenol Aの精子形成に及ぼす影響について検討を行った。
1.雄に対する直接的な影響
4週齢雄マウス(C57BL/6J)に200ng/kg/dayのBisphenol Aを週3回、6週間経口投与し、その後、20週齢まで経時的に観察した。その間、生殖器官重量および肉眼的観察による変化は認められず、妊孕性に異常はなかった。しかし組織観察により、精細管内精細胞の剥落、精細胞の壊死塊等が認められた。
2.母親を通じた間接的な影響
4週齢雌マウス(C57BL/6J)に200ng/kg/dayのBisphanol Aを週3回、6週間経口投与した後、交配し、さらに投与を続け、得られた新生仔の臓器重量及び、各臓器の肉眼観察を行った。その結果、多くの個体において精巣だけに、著しいサイズの減少が見られた。これらの異常精巣の組織像観察を行ったところ、その一部は精細胞分化障害により精子形成が完全に阻害されており、多くは部分的障害を示していた。さらにこれらの新生仔個体は10週齢まで、前立腺重量増加が顕著であり、これはエストロジェン類似化学物質であるDiethylstilbestrol(DES)投与により引き起こされる結果と同様の傾向を示した。
以上の結果は雄個体にBisphenol Aを投与した場合には直接精巣生殖細胞に対し、弱い影響が現れ、雌個体に投与した場合は、受胎後、Bisphenol Aの胎内暴露により、新生仔の精子形成阻害が引き起こされた可能性を示している。
