胎生および新生仔期内分泌かく乱物質の乳癌発生促進作用の機序解明

研究概要

我々はこれまでに、新生仔期の非常に強力な女性ホルモン作用を有する合成ホルモンdiethylstilbestrol(DES)がネズミにおいて、雄には精子無形成が、雌には無排卵状態などの内分泌撹乱を引き起し、乳癌の発生を促進することを明らかにした。本年度は、生活環境に実在し、DESの1000分の1程度しかホルモン作用のない4-tert-octylphenol(t-OP)と4-n-octylphenol(n-OP)でも同様の障害を惹き起こすかどうかを検索した。
1、カエルの生態観察を定期的に行っている田んぼで採取した卵をt-OPの水溶液で培養したところ、10ppm群では孵化せず、1ppmでは約10%は孵化したが、発育異常で孵化後14日目には死亡した。この結果より、OPがカエルの卵の孵化やオタマジャクシの生育を障害する可能性は否定できず、今後低用量群での長期観察が必要である。
2、SD系雄ラットに生後28、42、56日目に3回7,12-dimethylbenz(a)anthracene(DMBA)10mgを経口投与し、生後28日と70日目よりn-OPを餌に混ぜ投与したところ、非投与群と比べ、両群とも多数の乳癌が発生した。OPは大量であれば、乳癌発生を促進することが明らかとなった。
3、n-OPを雌ネズミへ生後24時間内に1回のみ皮下投与したが、内分泌やDMBAによる乳癌の誘発への影響はみられなかった。今後、ホルモン作用の極めて弱いOPは、形態形成時期でホルモンの影響をもっともうけやすい胎生期でも異常を惹き起こさないかどうかの検索が必要である。
これらの結果より、オクチルフェノール(OP)は単独で、短期間であれば、極めて大量でなければ、内分泌を撹乱しないように思われる。今後、多種類の環境ホルモンの暴露実験を行い、混合汚染の関与について検討を行う。
