プレT細胞に特異的に発現する新規遺伝子の解析

研究概要	RITは、未熟胸腺細胞の内でもCD4^-CD8^-DNのCD25+亜集団に最も高い発現が見られ、分化の進んだCD4^+CD8^+DP細胞ではもはや全く発現のない特異的な発現パターンを示す。そこで、2つの系で胸腺初期分化を調べた。pTαを発現するSCID由来のpre-T細胞株を刺激すると、pTα発現は下がり、TCRα発現が誘導される。この変化と共にRITの発現は消失する事が判明した。In vivoでの分化に伴う変化を調べるために、抗CD3抗体をRag欠損マウスに投与すると、TCR発現はないにもかかわらずDNからDPに分化する。この時にもRITの発現誘導が起こることが証明された。また、RITのGST融合蛋白を作製し、これを免疫してモノクローナル抗RIT抗体を樹立した。この抗RIT抗体を用いて細胞からの免疫沈降、細胞内分布を調べたところ、多くは細胞質に存在することが判明した。ロイシンジッパーを持つ分子構造から、核内の転写因子と考えていたが、その可能性もオープンにして解析する必要がある。In vivoでの機能を明らかにするために、RIT欠損マウスの樹立のために努力した。Targeting constructを作製し、homologous recombinationを持つESをスクリーニングし、これをblastcystにinjectionし、現在キメラマウスを数多く得ることができた。このマウスから必ずやgermline transmitされたKOマウスができ、分化における機能が解析できると信じる。一方、RITをレトロウイスルベクターを用いてFTOC(fetalthymus organ culture)の系に導入してin vitroでの分化における影響を調べる予定であったが、retrovirus vectorや package cell lineもいろいろ変えたものの感染と発現に成功せず、結論を出すに至らなかった。
