サイクロスポリンAとFK506の新たな標的

研究概要	免疫抑制剤であるサイクロスポリンA(CsA)やFK506はセリン・スレオニン脱リン酸化酵素であるカルシニューリン(Cn)の酵素活性を抑制する。従来Cnの抑制のみがCsAやFK506の作用機序と考えられてきた。我々はT細胞受容体とCD28の共受容体の刺激によって活性化されるMAPキナーゼファミリーのJNKやp38の活性化もCsAやFK506によって抑制されることを見いだした。一方、CsAやFK506は高浸透圧や薬剤によるストレスに伴って活性化されるJNKやp38は抑制しない。これらの事実はT細胞活性化に特異的なJNKやp38の活性化経路もCsAやFK506の標的であることを示唆する。そこで、この経路の分子的基盤を明らかにするための第一歩として、Cnの活性化経路を恒常的に活性化し、JNKやp38の活性化経路のみを検討できる系の開発しを試みた。そのために、Cnの基質でありかつT細胞活性化の最終標的遺伝子の一つであるインターロイキン2(IL-2)の転写に必須のNF-ATタンパク質の恒常活性型変異体を作製した。NF-ATはN端側にリン酸化を受けることによって細胞質内にとどまり、Cnによって脱リン酸化を受けると核内へ移行する。そこでリン酸化部位を含むN端側を欠失した変異体を作製したところ、変異体は恒常的に核内にとどまり、さらに転写活性も持つことが明らかになった。恒常活性型NF-ATを発現するT細胞に刺激を与えてIL-2プロモーターの活性化を検討したところ、やはりCsAやFK506がJNKやp38の活性化ならびにIL-2プロモーターの活性化を阻害することが明らかになった。これらの実験結果はCn/NF-AT経路とは独立にCsAやFK506の標的が存在することを強く示す。現在さらにその標的を同定すべく検討を重ねている。
