マクロファージにおけるインターロイキン18(IL-18)産生の制御機構

研究概要	IL-18は炎症・免疫反応において極めて重要な役割をしている。その作用はやや複雑でIFN-γのような強い炎症性サイトカインの誘導を通じて組織に損傷を引き起こす一方でIL-4,IL-13などの抗炎症性サイトカインを誘導する。このことはIL-18の産生とそのプロセシングの制御が重要な意味を持つことを示している。マクロファージは多量のIL-18前駆体を含んでおり、そのプロセシングは主にcaspase-1(ICE)によって起こると考えられている。しかしcaspase-1も前駆体の形で存在しており、多くの熱心な研究にもかかわらずその活性化の機序は不明のままである。in vivoの実験ではP.acnes処理マウスにLPSを投与すると30-50分の短いlag timeの後多量のIL-18が血中に遊離されるが、caspase-1遺伝子欠損マウスではそのようなことは起こらない。このことからIL-18を産生するマクロファージがP.acnes処理によって増加し、IL-18のプロセシングが急速なcaspase-1の活性化によって起こったと考えられた。このcaspase-1活性化の過程をブロックするかもしれないと考えられるようなantioxidantやtrypsin inhibitorなどをLPS投与直前に投与して調べたがIL-18の遊離を抑えることはできなかった。そこでP.acnesでelicitした腹腔マクロファージや肝クッパー細胞をin vitroでLPSその他の物質により刺激してみたが培養液へのIL-18遊離を起こすことはできなかった。しかしながら上皮系の細胞をIFN-γ、LPS、proteinase-3などで刺激すると多量のIL-18を遊離することが明らかになり(投稿中)、好中球由来のproteinase-3がなんらかの関与をする可能性が示された。現在このproteinase-3によるIL-18のプロセシングがcaspase-1の活性化を通じて起こるのかなどについて検討をしており、マクロファージによるIL-18の産生、プロセシングにも大きな示唆を与えるものと思われる。
