多因子疾患糖尿病における機能ゲノム学の新展開

研究概要	多因子疾患では複数の遺伝子と環境因子の相互作用により疾患の発症が規定されることから、責任遺伝子の同定・単離・機能解析には従来の単一遺伝子疾患に用いられてきたのとは異なる手法によるアプロチが必要である。本研究では、近交系モデル動物を用いるて遺伝的背景ならびに疾患を均一にするとともに、そのコンソミック系統・コンジェニック系統を作出して多因子を単一因子に機能分割することで、多因子疾患の原因遺伝子の同定・単離・機能解析を効率よく進める新たなアプローチ法を構築するとともに、ヒトのカウンターパートの解析を進めた。2型糖尿病の優れたモデル動物NSYマウスを対象に行ったゲノムスキャンで同定した耐糖能に強く影響する2つの遺伝子座(Nidd1n, Nidd2n)の存在する第11染色体、第14染色体をそれぞれ単独で有するコンソミックマウス(C3H. NSY-chr11, C3H.NSY. chr-14)を確立し、表現型を詳細に解析した結果、C3H. NSY-chr11はインスリン分泌低下およびインスリン抵抗性を示し・空腹時・負荷後血糖値がいずれも有意に高値を示した。C3H. NSY-chr14はインスリン抵抗性を示すが、インスリン分泌には変化を認めず、耐糖能も正常であった。いずれのコンソミックも親系統のNSYに認められるような肥満・内臓脂肪蓄積・レプチン抵抗性は示さなかった。以上の結果より、(1)Nidd1はインスリン分泌不全・抵抗性の両者を介して耐糖能に強く影響すること、(2)Nidd2は肥満とは独立してインスリン抵抗性をきたすこと、(3)Nidd2によるインスリン抵抗性はそれ単独では耐糖能は変化しないこと、が明らかとなり、多因子疾患の複雑な遺伝支配の機能分割による解明が大きく前進した。
